Bicycle Therapeutics is a clinical-stage pharmaceutical company developing a novel class of medicines, referred to as Bicycle® molecules, for diseases that are underserved by existing therapeutics. Bicycle molecules are fully synthetic short peptides constrained with small molecule scaffolds to form two loops that stabilize their structural geometry. This constraint facilitates target binding with high affinity and selectivity, making Bicycle molecules attractive candidates for drug development. The company is evaluating zelenectide pevedotin (formerly BT8009), a Bicycle® Toxin Conjugate (BTC®) targeting Nectin-4, a well-validated tumor antigen; BT5528, a BTC molecule targeting EphA2, a historically undruggable target; and BT7480, a Bicycle Tumor-Targeted Immune Cell Agonist® (Bicycle TICA®) targeting Nectin-4 and agonizing CD137, in company-sponsored clinical trials. Additionally, the company is developing Bicycle® Radio Conjugates (BRC™) for radiopharmaceutical use and, through various partnerships, is exploring the use of Bicycle® technology to develop therapies for diseases beyond oncology.

Job Description
The Senior Director of Drug Metabolism and Pharmacokinetics (DMPK) reports to the Vice President of Quantitative Pharmacology group. In partnership with drug discovery medicinal chemists, biologists, pharmacologists and toxicologists, you will be responsible for leading a small team of DMPK scientists supporting multiple projects from target identification through to candidate selection and ultimately regulatory submission.
- Provide DMPK scientific leadership across all stages of the portfolio.
- Oversee the quality and performance of all DMPK activities including interactions with external CRO’s and internal project stakeholders.
- Work collaboratively with other core disciplines to support compound progression based on optimizing DMPK properties.
- Integrate data from multiple sources to inform on the understanding of bicyclic peptide distribution, metabolism and excretion.
- Work closely with in-vivo pharmacology to design experiments to understand preclinical pharmacokinetic/pharmacodynamic (pk/pd) relationships.
- Work closely with discovery team and regulatory team to author and review DMPK related IND documents
- Managing DMPK budgets for CRO’s and monitoring CROs to ensure delivery of quality data on time.
- Support leadership of the Quantitative Pharmacology group and junior staff development
